Two Numbers Get Cited for Cagrilintide, and Only One Describes What’s in the Vial
Cagrilintide is a long-acting amylin analog, engineered to hold the same satiety and gastric-emptying signal the body’s own amylin sends after a meal, but for a week instead of minutes. That is why the trials below dosed it once weekly. The confusion is not about the compound. It is about which trial a given number actually belongs to, and Etched carries cagrilintide as a standalone compound, not the combination most headline figures come from.
Where the Higher Number Comes From
The most-cited cagrilintide weight-loss figures come from a phase 1b trial that paired cagrilintide with semaglutide 2.4mg, not cagrilintide on its own (Enebo et al., Lancet, 2021, PMID 33894838). At week 20, the combination arms reported roughly 15.7% and 17.1% mean body-weight reduction. Those numbers describe the pair. They do not describe cagrilintide by itself, and citing them as if they do misattributes someone else’s result to this material.
The Number That Actually Matches What Ships
The standalone trial is a separate, larger phase 2 dose-finding study (Lau et al., Lancet, 2021, PMID 34798060, registered as NCT03856047), dosing cagrilintide alone across a 0.3mg to 4.5mg range for 26 weeks. Its reported reductions ran roughly 6.0% to 10.8%, real numbers, and meaningfully lower than the combination figures above. That is the study whose population and dosing actually match a standalone cagrilintide vial. Why the Gap Exists
Amylin and GLP-1 act on different receptor pathways, which is exactly why some trials paired them: to test whether hitting both signals at once produced more than either alone. It did, in that trial. But that finding belongs to the combination, and pairing compounds is a separate research question from what a single compound does by itself. A standalone amylin signal and a combined amylin-plus-GLP-1 signal are not the same experiment, and the literature should not be read as if they were.
What This Does Not Establish
Cagrilintide is investigational. It carries no approval for any use, and nothing here says it is effective or safe in general use. Both trials cited are phase 1 and phase 2, early to mid-stage, run in specific populations for 26 weeks or less. Neither establishes long-term outcomes or results in the general public.
